t ¥ August 17, 1988 SCIENCE Alzheimer’s linked to virus TORONTO (CP) — Some cases of Alzheimer's disease may be caused by an unidentified, slow-acting virus, say researchers at Yale University in Princeton, N.J. A team led by Dr. Elias Manuel idis, a neuropathologist at tha Yale school of medicine, say its exper. iments are the first in 20 years to find a link between an infectious agent and Alzheimer's. But they stress their findings are tentative and need to be confirmed. Alzheimer's causes its victims to lose their memory and suffer de- mentia, as it slowly destroys their minds and personalities. More than 2.5 million North Americans are believed to suffer from the disease. Manuelidis attributes the mental disorder to a rare blood-borne virus. The virus produces changes in brain cells resembling a rare, progressive disorder called Creutzfeldt-Jakob disease. Although the disease re- sembles Alzheimer’s, it progresses differently in the brain. Manuelidis suggests a genetic element, possibly triggered by exp- osure to the slow virus, may cause people to be susceptible to Al zheimer’s. The Yale researchers, who pub- lished their findings in the July issue of the Proceedings of the National Academy of Sciences, took human blood cells from 11 volunteers and injected them in the brains of hamsters. CHECKS VIRUS They were trying to test the infectious nature of what they believed was a virus causing the disease. In a release from the National Institute on Aging, Manuelidis said all previous attempts to transmit Alzheimer's failed because research- ers were using tissue samples containing a weakened virus from people in the final stages of the disease. So the researchers sought samples from people who might be in the early stages of the disease. All 11 volunteers had at least two close relatives with Alzheimer's. Nine of them had no evidence of the disease. Two had mild signs of mental disorder and later developed Alzheimer's White blood cells from the vol unteers samples were then injected into hamsters, which were observed for signs of the disease. Within one year, cells from five of the volunteers resulted in brain degeneration in the hamsters. One animal was confirmed as having Alzheimer's. A second group of hamsters was inoculated with brain cells taken from the five infected hamsters. All hamsters in the second group dev eloped a more severe form of the disease. But experts stress these exper iments don't indicate Alzheimer's is transmitted by person-to-person con tact. Even if a virus is eventually linked to the disease, research has found no evidence Alzheimer's is contagious. Gurgles mean something REGINA (CP) — The giggles, gurgles and grins of a toddler with speech problems are building blocks for communication, says a speech- language pathologist. Parents who respond to those attempts to communicate can help their children learn, says Judy Bell, who works for the Lakehead board of education in Thunder Bay, Ont. “We often miss these in chil dren,’’ Ball said of the eye contact, gestures and body movement used by children with speech or language problems. ‘‘By the time we tune in, they're gone.”’ Ball was in Regina for a recent three-day workshop by the Saskat- chewan Association of Speech Lan- guage Pathologists and Audiologists. The program, developed 14 years ago in Montreal, is based on the notion that children with language problems get caught in a vicious cycle because their parents respond to their level of communication. x Good Business $sense... (AP) — Studies in diabetic mice suggest that a drug that stimulates the immune system may stop the destruction of insulin-producing cells and thus prevent diabetes, a researcher said. Stress and viral infections also have been found to affect the development of diabetes, said Ed- ward Leiter, a geneticist at The Jackson Laboratory in Bar Harbor. Leiter said he and his colleagues id expected: that an immune- stimulating drug would make dia- betes worse in the mice they studied. Instead, it apparently brought the diabetes under control. The form of diabetes that requires insulin injections is thought to occur when the body's immune system attack its own insulin-producing cells, or beta cells, in the pancreas, Leiter explained. The drug, a substance called interleukin-2 that occurs normally in the body in small amounts, activates the immune system cells and thus should have accelerated the des. truction of beta cells, Leiter said. It appears, however, that the drug activated some kind of immune cell that has the normal function of preventing the body's defences from attacking the body itself. In the mice, it blocked the destruction of beta cells. This finding — in mice specially bred to have diabetes — may not hold any immediate importance for hu- mans, but it does shed light on the in ithe males of that mouse strain. male mice left alone in a basic processes ig the dev- elopment of diabetes, Leiter said. Mice and humans share many genes and a variety of diseases. SEPARATE GENES Leiter has determined that in the mice he studied, a strain called non-obese diabetic or NOD, at least three separate genes seem related to the development of diabetes. He has also found that stress appears to aggravate the develop- ment of diabetes, which occurs only cage with a female for breeding developed diabetes at about half that rate, Leiter said. Leiter said that Michael Oldstone of the Scripps Clinic in La ‘Jolla, Calif., recently found that infection with a virus could block the destruct- ion of beta cells in NOD mice. Leiter said viral infections stim- ulate the production of interleukin-2. Oldstone's infected mice might have produced more interleukin-2.
